Abstract
A sensitive, selective, and precise RP-HPTLC method has been developed for the quantification of podophyllotoxin in Podophyllum hexandrum, collected from different locations of the western Himalaya of India, ranging from 3250 to 4100 meters above the mean sea level. The assay employed RP-18 F-254 TLC plates (chromatoplates) as the stationary phase. The solvent system consisted of acetonitrile-water (4: 6, v/v) and gave better resolution, with well separated, compact spots for podophyllotoxin (Rf value 0.41 +/- 0.02). Densitometric analysis was carried out in the absorption remission mode at 217 nm.
